Nester, Eugene W. (Western Reserve University, Cleveland, Ohio) and John Spizizen. Role of one-carbon precursors in the biosynthesis of deoxyribonucleic acid in bacteriophage-infected and growing cells of Escherichia coli. J. Bacteriol. 82:867-874. 1961.-The ability of growing and T2 bacteriophage-infected cells of Escherichia coli to incorporate serine-3-C(14), glycine-2-C(14), formate-C(14), and formaldehyde-C(14) into purine and pyrimidine moieties of deoxyribonucleic acid (DNA) was determined. All four one-carbon precursors are effective contributors to the DNA-purines, but only glycine-2-C(14) and serine-3-C(14) are incorporated into the side chains of the pyrimidines. In addition, formate-C(14) becomes incorporated only into position 8 of the purine ring, whereas isotope from serine-3-C(14) and glycine-2-C(14) is incorporated equally into the 2 and 8 positions. No qualitative differences were observed in the patterns of incorporation of any one-carbon units in growing or bacteriophage-infected cells. However, the 3-carbon of serine serves as a more effective precursor of the 2 and 8 positions of the DNA purine ring when the cells are infected. Formate-C(14) and to a slight extent glycine-2-C(14) are somewhat better precursors of these positions when the cells are infected under appropriate conditions.

Isolation of lytic bacteriophages against pathogenic Escherichia coli strains in poultry in the northwest of Iran
In this study, 90 internal organ samples of poultry with symptoms of colibacillosis were obtained from Maragheh poultry farms in East Azerbaijan, Iran. In total, 70 bacterial isolates were confirmed as Escherichia coli (E. coli) strains using standard biochemical tests, and antibiotic sensitivity was determined by the disk diffusion method. Antibiotics used in this study included ampicillin, pe...
متن کاملEffect of Concomitant Lycopene Biosynthesis on CoQ10 Accumulation in Transformed Escherichia coli Strains
CoQ10 and lycopene are isoprenoid compounds with nutraceutical and pharmaceutical benefits. In this study, the effect of concomitant lycopene biosynthesis on CoQ10 accumulation in transformed Escherichia coli DH5α was studied. A lycopene production pathway including geranylgeranyl diphosphate synthase (crtE), phytoene synthase (crtB), and phytoene desaturase (crtI) from Erwinia herbicola was co...
متن کاملThe origin of purine and pyrimidine deoxyribose in T6r+ bacteriophage.
Lanning and Cohen (1) reported that when Escherichia coli strain B is infected with T6r+ bacteriophage in a medium containing glucose-1-Cr4, the deoxyribose from the deoxyribonucleic acid of the T6rf particles liberated on lysis of the cells contains 40 to 60 per cent of the specific activity of the exogenous glucose. In parallel experiments, it was shown that the deoxyribose from the deoxyribo...
متن کاملExpression of Recombinant Protein B Subunit Pili from Vibrio Cholera
Background & Aims: Vibrio cholerae is a gram-negative bacterial pathogen that causes cholera disease. Following ingestion by a host and entry into the upper intestine, V. cholera colonizes and begins to emit enterotoxin. One of the most pathogenic factors of Vibrio cholera is toxin-coregulated pili (TCP). ToxinCoregulated pili is as the primary factor requiered for the colonization and insisten...
متن کاملThe Effect of Chloramphenicol on Deoxyribonucleic Acid Synthesis and the Development of Resistance to Ultraviolet Irradiation in E. Coli Infected with Bacteriophage T2 by Jun-ichi Tomizawa
In spite of its capital importance, the mechanism of deoxyribonucleic acid (DNA) synthesis remains quite obscure. One of the main reasons for this is the complicated correlations between the synthesis of DNA and other cellular constituents in cells. In phage-infected bacteria the metabolic pathways shunt to phage formation, resulting in production mainly of DNA and protein. Thus the system is c...
